Cdk5 Sebagai Target Potensial Penemuan Obat Anti Nyeri Kronik

Abstrak

Nyeri kronik merupakan suatu kondisi yang sangat mengganggu dan hingga saat ini belum ditemukan obat yang efektif dan aman. Hal ini bisa disebabkan karena obat yang ada belum mengenai target penting dalam transmisi nyeri kronik. Cdk5 adalah protein kinase yang telah terbukti memiliki peran kunci dalam transmisi nyeri kronik. Peran penting tersebut antara lain :  pengaktifan TRPV1 ,NMDA, P/Q type voltage- dependent calcium channel, dan reseptor ATP-gate P2X . Cdk5 bisa dijadikan sebagai target potensial dalam pengembangan obat anti nyeri kronik yang baru.  Beberapa aspek yang menjadi perhatian dalam pengembangan obat yang mentarget Cdk5 tersebut adalah peran Cdk5 dalam kondisi fisiologis dan patologis karena hal ini akan berhubungan dengan resiko efek samping. faktor lain yang juga sangat perlu dipertimbangkan adalah lokasi ekspresi Cdk5 karena hal ini akan sangat menentukan desain obat. Beberapa obat yang telah dikembangkan sebagai penghambat Cdk5 adalah roscovitin, olomoucine, dan indirubin.
